Yes, huge differences. What are you trying to accomplish, we can probably point you toward which input you will want to use.

i want to make zone that this robot can enter only after getting signal from PLC that other robot is in HOME position and im not sure how i can do that properly

We are going through this ourselves right now. First, do you know if you have the Ethernet Safety option, or the Devicenet Safety Option?
Also, what controller and cabinet?

If you have Ethernet Safety or DeviceNet Safety, you will want to use CSI, if not, you will be using SPI. SPI is a hard wired connection on the E-Stop board inside the cabinet (this is what we are working on doing right now). We have R-30iB A Cabinet. The A-Cabinet only has two inputs and two outputs. B Cabinet has more.

Read your DCS manual, it has a table that goes over what each signal is, and what drives them.
Is this related to your other post where both arms are on the same controller? If that is the case, define a joint check where the one arm is at home, and then mute the zone with the JPC. You may have to invert the signal using the safe io, and an internal safety relay bit (SIR)
Also, machine level safety (i.e. the only thing that could get damaged is a portion of the machine) doesn't require safety level signals. Aka, you shouldn't do this in DCS. Use a reference position on the other first robot instead. In my opinion, DCS should only be used in cases where a human could get hurt or killed.

i got profinet safety i/o so CSI, R-30iB, as i see cabinet B, also i see in DCS SAFE I/O device type E-stop board with 8 SPI /8 SPO
its not directly related with my other post, those 2 (welding) robots on one controller stay near robot with gripper with will pass tool.
i want to be sure that gripper wont come, when welding robots are in work position
