IRB6640 manual says I have to lockout/tagout before replace the SMB battery. I wonder if the manual is saying that for a legal reason. I am used to Fanuc robots where I make sure I do NOT remove power while I change out SMB batteries. I do not want some freak event happen and I do not replace the batteries fast enough (or the batteries were faulty) and end up losing the calibration data. If I have the power on, at least the battery is out of equation.

I prefer to change batteries with power on... but if LOTO insists, I drive robot to calibrate position before powering down (usually 0,0,0,0,0,0 but we have a few painters that reference elsewhere.) Yes, robot will lose rev counters, but it is already where it needs to be for a reset. This is on an S4(C/+) system. I haven't had the opportunity to change SMB batteries on an IRC5 yet.

It does not matter as long as you move the robot to the zero position before turning robot off.
When you turn off it will lost revolution counter, but as you already put the robot in zero position, it's just a matter of updating.
